In our Biobank we have about 100 000 tissue samples (99.9% ethanol) and DNA extracts stored altogether stored in -80 freezers. Each tissue sample has a label with the tube number inside and every DNA extract has a sticker with a number outside the tube. These numbers are not always unique but be that as it may, these samples have unique freezer locations and are linked to Collection objects in the Specify database. In the quest to improve how we store and label DNA extracts and tissue samples, I am curious to find out from colleagues who have Biobanks in their museums as what they consider to be best practice. I would direct you to the SPNHC best practices wiki page on Genetic Resources for the most comprehensive best practices on tissue collections - https://spnhc.biowikifarm.net/wiki/Genetic_Resources. There is a wealth of information here that should help. In general in response to your request for info on storing and labeling I have always steered clear of putting labels into cryo tubes for fear of contamination and rely instead on cryo labels affixed to the outside of the cryotubes. There are great labels available that will stick to already frozen tubes and include barcodes and human-readable numbers. It would be ideal to have unique catalog numbers associated with each tissue but I realize that this is sometimes just not possible with a collection that large. I would also encourage you to titrate off all ethanol in the collection as exposure to ethanol will slowly degrade the tissue quality at -80 temperatures. We maintain all of our tissues dry in cryogenic dewars with all liquid (ethanol, DMSO, RNA Later) having been poured off before final storage. Hope that helps. But using spirits of wine, He found they did fine, Which we celebrate each 4th of June. Old Croone Day is an international holiday to celebrate the first recorded mention of the preservation of scientific specimens in alcohol, which took place on 04 June of 1662. On that momentous occasion, Dr. William Croone (1633-1684) showed his fellow members of the Royal Society of London ?two embryos of puppy-dogs, which he had kept eight days, and were put in spirit in a glass-vial sealed hermetically.? Croone was a founding member and secretary of the *Royal Society of London for Improving Natural Knowledge*, the oldest scientific society in the world. Ironically, his name was misspelled as Croune in the meeting records (see below). Croone, a graduate of Cambridge University (Emmanuel College), variously served as a professor of Rhetoric at Gresham College, a physician, and an anatomy lecturer while pursing diverse interests in physics, physiology, and embryology. It was Croone?s interest in entomology that led him to discover the possibilities of fluid preservation in spirits of wine (ethyl alcohol). While most of my skeleton work has been many thousands of birds and herps, the few hundred mammals I have worked on show some idiosyncrasies. Mammal teeth have a much harder outside layer and a softer inside. If you try to dry the teeth very fast in a non-humid environment, most pf the time canines and many other teeth will crack vertically. I will often slowly dry the skulls when teeth are intact within a jar with the lid partially open. If the time to dry is spread out over a couple to a few days, the likelihood of cracking is reduced - but never guaranteed. I have also seen cracked major long bones on even larger mammals like the North American White-tailed Deer. My area is not particularly dry like Israel may be, so I would use caution drying mammal skeletons quickly. Bird skeletons, as well as reptiles usually aren't as thick walled, and I have not seen the cracked bones in these groups. Stephen P. This is because we did not want the added calcium carbonate in the box and we also are worried the buffer might fail at some point. However, it is very hard to find a un-buffered boxes of a wide variety of sizes. We looked into UniversityProducts, Gaylord and Talas as suppliers but none had a good selection of un-buffered boxes. Our first question is how important is using un-buffered boxes vs. buffered ones? The second is: Does anyone have a name for a good supplier of un-buffered boxes that have a good selection of sizes? The remainder we put in a small glassine envelope which is also numbered. Currently our skeleton numbers are 5 digits. We use a fine Rapidograph pen with India ink. I guess the question is how will your specimen be used? Our specimens are often used in side by side comparison so the chances of mixing are high. I often find bones in the wrong box but am able to reassociate them because they are numbered. However, we do not number them by writing directly on the bone - this creates situations where mistakes can't be corrected in a good way and where changes in the numbering system (which may, and do, occur in many collections over the years) result in writing a new number alongside a pre-existing one - or pre-existing ones in plural! - rather than taking off the old number (which is preserved in the relevant specimen record in the catalogue book and in the database). Instead, we apply a film of Paraloid B72 (dissolved in acetone) to the bone and write on the film - not on the bone itself. Thus, it is possible to remove or correct numbers. PB72 is a well-known material in conservation and is supposed to be durable and stable enough to be of good use in numbering specimens. Yes, all this still requires a lot of work, but this is part of making sure that skeletal parts do not get mixed up, while taking into consideration any changes that may happen in the future. This includes very small bones (hummingbirds, shrews, etc.). We use a fine archival pen for numbering. In the event of a numbering mistake, we gently scrape off the number with a razor blade and renumber. This work is done mostly by undergraduates, and we ask for a sample of their handwriting in advance to make sure that they can write small and legibly. Schmidt, the Zoological Collections Manager for the Sternberg Museum of Natural History at Fort Hays State University. Curtis became the Museum?s first official Zoological Collections Manager in July of 2011. A Hays native and FHSU grad, Curtis received his BS in biological sciences in 1999 and his MS in 2004. Curtis was no stranger to the museum, as he began as a curatorial assistant in Herpetology in 1998, eventually becoming an associate curator of Herpetology and Mammalogy before being appointed Zoological Collections Manager. I did some research there and they have 2 Boxes full of very interesting correspondence with Karl M?bius including some cataloges and price lists of specimens (handwritten and printed). Linnaea in fact was established two times in the Invalidenstrasse and was very important for the first display collections of MfN in Berlin. In-Reply-To: References: Message-ID: Hi all, Belated follow-up to my rant below, just in case it helps anyone else. Even after our graduate RA spent hours over multiple weeks with Honeywell customer service and resolved none of our problems, our new IT guy sat down with our machine for 2 hours today and fixed nearly every issue!! If anyone wants advice on this machine, I can probably put you in touch with him directly (I myself was not involved in the process). Most were easy built-in settings, once he found the right menus on the machine (not in the program we use to print). See edits to my original text, below each in red. 1. Connecting it to a device was harder than it should have been (our in-house IT could not do it without Honeywell customer service help). 2. It prints very faintly. We have it turned up to maximum density and it is just barely at the darkness our old Datamax was on a regular setting (and we have been informed by Honeywell that printing at darkest setting with cause it to die out faster?). * This was fixed by adjusting the heat output settings, NOT the darkness setting. 3. It doesn?t seem to recognize the media we already had (supposedly ?standard? and can fit any thermal printer). This results in us having to open and shut the print head before every label, just to trick it into recognizing there is media present. It also ejects/feeds about 4-5 labels worth of media after each label. * While we may still not be using the preferred media, he got it to at least stop thinking it was ?out of media? by turning off the sensor that is at the front of the machine, just where the label pops out. * The second half of this issue was fixed by changing the setting for the type of media. It now prints continuously with no issue. 4. There is no built-in cutter to cut your labels for you (not sure if that is just a model issue or none of these Honeywell ones have it; the company seemed confused by this concept) 5. We use FileMaker as our specimen database and our presets on labels there print ok, but the ones in Excel do not (and we basically have to do weird re-formatting things in Excel every time we create a new label) * He is now working on pre-sets now to hopefully avoid this issue too! We also requested the ?official? Honeywell media sample thinking maybe it would start to recognize it (perhaps fixing multiple issues in #3). They have not sent the media yet (requested over a month ago), so I assume we will have to eventually break down and just buy it ourselves, hoping it fixes the problem. * Honeywell customer service is clearly not great; we have still only received the ?black? ribbon not the ?white? so we can?t really try out the ?preferred? media yet (even though the request was nearly 2 months ago). Additionally, the roll they sent is NOT 4? wide which is what all our labels are set for? Best of luck in your own printing endeavors! We bought the following machine last Fall: Honeywell PX940 https://www.cdwg.com/product/honeywell-px940-3-core-300-dpi-thermal-industrial-printer/5712690?pfm=srh It has been a total disaster and we all feel like we wasted a few thousand dollars. Details are as follows: 1. Connecting it to a device was harder than it should have been (our in-house IT could not do it without Honeywell customer service help). 2. It prints very faintly. We have it turned up to maximum density and it is just barely at the darkness our old Datamax was on a regular setting (and we have been informed by Honeywell that printing at darkest setting with cause it to die out faster?) 3. It doesn?t seem to recognize the media we already had (supposedly ?standard? and can fit any thermal printer). This results in us having to open and shut the print head before every label, just to trick it into recognizing there is media present. It also ejects/feeds about 4-5 labels worth of media after each label. 4. There is no built-in cutter to cut your labels for you (not sure if that is just a model issue or none of these Honeywell ones have it; the company seemed confused by this concept) 5. We use FileMaker as our specimen database and our presets on labels there print ok, but the ones in Excel do not (and we basically have to do weird re-formatting things in Excel every time we create a new label) We have tried a new printhead, thinking it would fix the faintness issue (#2), but it did not. We also requested the ?official? Honeywell media sample thinking maybe it would start to recognize it (perhaps fixing multiple issues in #3). They have not sent the media yet (requested over a month ago), so I assume we will have to eventually break down and just buy it ourselves, hoping it fixes the problem. Hello, I would like to know if anyone has experience using the following thermal label printer for collections stored in ethanol: HONEYWELL, PX940 PRINTER, VERIFIER W/PERPETUAL LIC., FULL TOUCH DISPLAY, UNIVERSAL FW, ETH, USB, SERIAL, LOW POWER BLUETOOTH, RIBBON INK IN/OUT, MEDIA CORE 3 INCH, DT AND TT, 300 DPI, US POWER CORD. Our efforts to purchase a Datamax or CAB/Squix were unsuccessful last year because of supply issues so we purchased a Honeywell option. We returned it to Honeywell in January because it was defective and we have now been offered the option to replace it with a slightly different model, the HONEYWELL, PX940 PRINTER, VERIFIER W/PERPETUAL LIC. I am also curious if anyone knows of a museum service available to have thermal labels printed by an external organization using the appropriate media and SDR ribbon? I don?t want to run the risk of ?Alphabet soup? in the jars of ethanol caused but incorrect ribbon. We need to deal with a large backlog of labels to be printed that have been accumulating over the past 2 years. As always, I really appreciate your advice.
