PET neuroimaging is a long-established technology to measure brain receptors, transporters, and enzymes in humans and animal models using a wide variety of targeted radiopharmaceuticals. The Yale PET Center is a world leader in PET neuroimaging, having developed and applied over 50 radiopharmaceuticals in human studies of healthy controls and a wide range of neuropsychiatric conditions. This presentation will start with an introduction to quantitative PET imaging with examples of imaging agents relevant to a wide variety of neurotransmitter systems. A major development pioneered at Yale was synaptic imaging, with novel PET radiopharmaceuticals which bind selectively to a synaptic vesicle protein, SV2A, a target that is ubiquitously expressed in the brain. The presentation will include the development of these ligands and recent clinical studies in Alzheimer?s disease and Parkinson?s disease. SV2A-PET opens the opportunity for a novel imaging biomarker of neurodegeneration and synapse-modifying treatments to complement amyloid and tau imaging. The presentation will also include recent imaging results from a next-generation dedicated brain PET system, the NeuroEXPLORER, developed as part of a BRAIN Initiative grant (U01EB029811), which provides the highest resolution human brain PET images ever achieved.
