This supersedes an earlier demo plan that Leica has decided to cancel for various reasons -- we apologize for any confusion this may have caused. The demo unit is a versatile confocal system featuring White Light Lasers and the latest hybrid detectors, along with several advanced capabilities: * FALCON FLIM system: capable of real-time FLIM and FLIM-FRET, a dye separation mechanism based on fluorescence lifetime. If you are familiar with the TauSense module on CCMI's Stellaris, FALCON builds on it with expanded functionality. The only other FALCON system at Yale currently is in the West Campus Imaging Core. * SpectraPlex module: supports separation of up to 15 colors, offering potential for 3D spatial biology applications. No other system with SpectraPlex is available at Yale. We encourage you to take advantage of this opportunity and sign up for a demo slot. While testing FALCON or SpectraPlex requires well-prepared samples that might not be ready in time, bringing any sample to the demo will help evaluate the system's general performance. This demo marks the final stage of our evaluation of cutting-edge point-scanning confocal systems, a process that we began late last year with the goal of expanding our imaging capacity and capabilities in this category of instruments. We plan to make a choice soon after this demo. Thank you for your participation and here again is the link to the sign-up sheet. For any technical questions, please contact Albert Ayoub (Albert.Ayoub at leica-microsystems.com) of Leica. To what extent are these abilities driven by shallow heuristics vs. deeper abstractions? I will argue that, to answer this question, we must view LLMs through the lens of generalization. That is, we should consider the data that LLMs were trained on so that we can identify whether and how their abilities go beyond their training data. In the analyses of LLMs that I will discuss, this perspective reveals both impressive strengths and surprising limitations. For instance, LLMs often produce sentence structures that are well-formed but that never appeared in their training data, yet they also struggle on some seemingly simple algorithmic tasks (e.g., decoding simple ciphers) in ways that are well-explained by training data statistics. In sum, to understand what AI systems are, we must understand what we have trained them to be. Speaker bio: Tom McCoy is an Assistant Professor of Linguistics at Yale University, with a secondary appointment in Computer Science. His research aims to bridge the divide between linguistics and artificial intelligence: how can we create AI systems that replicate the rapid learning and robust generalization that humans display when processing language? Much of this work involves analyzing the performance and internal processing of neural network language models. He received his PhD from the Department of Cognitive Science at Johns Hopkins, and his PhD thesis received a Glushko Dissertation Prize from the Cognitive Science Society. He then did a postdoc in Computer Science at Princeton before joining the faculty at Yale. Outside of research, he is an organizer and problem writer for NACLO, a contest that introduces high school students to linguistics and natural language processing. Website. Warm regards, Paras Patel, PhD On behalf of the Yale Neuroscience Post-Doctoral Committee Title: When your Ferrari breaks down: Metabolic-functional coupling of fast-spiking parvalbumin inhibitory neurons in the healthy and injured brain Abstract: Parvalbumin-positive interneurons (PV-INs) are critical regulators of circuit activity and are selectively vulnerable across neurological disorders, including traumatic brain injury (TBI), epilepsy, Alzheimer?s disease, and schizophrenia. Their susceptibility is thought to stem from high metabolic demands driven by intense electrical activity. Using single-nucleus RNA-sequencing (snRNAseq) from rodent and human TBI, we show that PV-INs possess unique metabolic specializations that are lost after injury but can be restored in vivo by the glycolytic inhibitor 2-deoxyglucose. We define a PV-IN transcriptional identity module?encompassing ion channels, metabolic enzymes, and synaptic machinery?that reveals heterogeneous subsets of injury-associated PV-INs with disrupted identity. Loss of metabolic specialization is tightly coupled to this transcriptional dysfunction, with the PV-IN?enriched transcriptional co-activator Ppargc1a emerging as a key driver. We further identify a family of long non-coding RNAs enriched in dysfunctional PV-INs that inversely correlates with metabolic specialization. Finally, analysis of published human TBI snRNAseq datasets confirms nearly identical PV-IN changes, underscoring the central role of PV-IN metabolic dysfunction in TBI pathology. There are still plenty of demo slots available. Please see details below. Thank you for your participation. For any technical questions, please contact Albert Ayoub (Albert.Ayoub at leica-microsystems.com) of Leica.
