Dirk,

I vaguely recall a thread from several years ago about this very topic.  Someone had done some analysis and talked to the tube manufacturer and their conclusion was that the silicone gaskets did not seal in ethanol.  A search of the nh-coll archives might turn it up as I am clearly a bit vague on the details.  They did an assessment of a variety of tubes and recommended the Nalgene ones. We specifically selected ungasketed, externally threaded tubes even for -80 freezers which we were using at the time.  There were too many reports of gasket failure at low temps and externally threaded tubes, although taking up slightly more space in storage, negate any contamination issues that may occur by having an internally threaded closure potentially in contact with tissue or preservation fluid.

Hi all,

I would like to know if there are any specific experiences (good and bad) on DNA-storage tubes (normal freezing as we don't have liquid-N storage).

We are using Matrix racks (sepra seals) and I want to change to high quality skrew-lid tubes, but I am sceptical with the silicone gasket in combination with the internal thread of the tubes.

Thermo says that the tubes are tight sealing (from liquid-N to normal freezing and even room temperature), but if the gasket starts to crumble, then the tubes necks are open. Also, gaskets are made of silicone, which might cause issues with cold-flowing behaviour and life span of the material.

Curious to hear your thoughts, especially if you have experiences where things turned out to be not that good as originally promised by the manufacturer (also off-line, if you wouldn't like to post any failures to the broad public)
